If I’m in a pub and I hear a track that I like and go on to the iTunes Store and buy that track. Is the mp3 file download to the music app on my MacBook the next time I sync.

It should. Or, you can simply download it again in the iTunes Store. Or, if you use iTunes Match or subscribe to Apple Music, the file will sync through the cloud.
