No. Buying more iCloud storage has no effect on your iPhone's local content. It will not delete anything on the iPhone.

It will also not affect your iPhone's local capacity in any way. iCloud Storage is on an Apple server somewhere. When you rent more space for the monthly fee, it will allow anything that has not been backed up due to space constraints there, to be new be backed up in the new available space.

It will not delete anything from iCloud either, it will just increase the available storage space there.
